The best-known serine/threonine kinases of the cyclin-dependent kinase (CDK) family regulate transitions between different phases of the cell cycle. However, many CDKs are involved in transcription but not in cell cycle progression. Among these “transcriptional CDKs”, CDK8 and its closely related paralog CDK19 play a unique gene context-specific role in transcription. CDK8/19 interact with different transcription factors and selectively potentiate the induction of previously silent genes, thereby acting as mediators of transcriptional reprogramming. Due to this unique function, selective small-molecule inhibitors of CDK8/19 are not toxic to mammalian cells or tissues under homeostatic conditions but have major effects in preventing biological reprogramming in different pathological and physiological processes. The effects of CDK8/19 inhibitors will be discussed in oncological settings, in particular in relation to cancer metastasis and drug resistance.
