The method for genetic transformation of different sugar beet varieties

A method for the genetic transformation of sugar beet (Beta vulgaris L.) by a binary vector LBA4404 (pAL4404, pGA482) of Agrobacterium tumefaciens was developed. Regenerated plants growing on a selective medium with kanamycin were obtained by inoculating leaf petioles with agrobacteria. The transgenic nature of kanamycin-resistant regenerated plants was proven by the presence of neomycin phosphotransferase activity and the respective gene npt II in their cells. The frequency of transformation depended on the induction of agrobacterial vir-function, the sugar beet variety, the type of explant, and the conditions for the transformant selection. The frequencies of transformation for different beet varieties by the wild strains of A. tumefaciens and A. rhizogenes were compared. Due to the high sensitivity of sugar beet to the wild strains of agrobacteria, the construction of new effective agrobacterial vectors for genetic engineering seems feasible.
