A novel water-soluble substrate for silicateins

It has been proposed to use tetrakis(2-hydroxyethyl) orthosilicate (THEOS) as a substrate for silicateins, a family of enzymes that play a key role in the formation of the marine sponge skeleton. A comparison of THEOS with the currently widely used tetraethyl orthosilicate (TEOS) has been carried out in the reaction of the formation of amorphous silica in vitro, catalyzed by silicatein A1 from the sponge Latrunculia oparinae. It has been shown that the reaction with THEOS proceeds much more intensively than with TEOS, probably due to a high water solubility and a higher hydrolysis rate of THEOS.
