Plant defensins as potential new antimycotics acting against sensitive and resistant strains of Candida albicans
Recently, fungal infections have become a serious clinical problem. One of the fungal pathogens of critical priority according to WHO data for 2022 is Candida albicans. C. albicans is a normal commensal that causes opportunistic infections in immunocompromised patients. C. albicans is a common cause of oropharyngeal and vulvovaginal candidiasis, skin and mucosal candidiasis, and, in more serious cases, systemic infections such as candidemia and invasive candidiasis, which are accompanied by damage to internal organs and are characterized by high mortality. Patients with chronic lung diseases, tuberculosis and diabetes, HIV-infected, cancer patients, premature babies are at risk. According to minimal estimates, annually invasive candidiasis is diagnosed in 250 thousand patients. At the same time, mortality from invasive candidiasis caused by C. albicans, despite ongoing antifungal therapy, ranges from 20% to 50%. The main reasons for this are the limited range of antimycotics used in clinical practice and the spread of C. albicans strains with cross- and multiple-resistance. Therefore, the search for new drugs for the treatment of superficial and systemic candidiasis, including those caused by resistant strains of the fungus, is especially relevant. Prototypes of such drugs can be plant defensins, which have a pronounced antifungal effect, high resistance, low toxicity, and immunomodulatory properties. This project is aimed at solving a fundamental scientific problem - the study of plant defensins as potentially new antimycotics that act against susceptible and resistant strains of Candida albicans. The applied significance of the project is due to the possibility of developing new drugs for the treatment of candidiasis, including those caused by resistant strains of Candida albicans.
