Analysis of changes in chaperone-dependent proteostasis and autophagy in the cells of the immune system during the development of Parkinson's disease

Parkinson's disease (PD) is a neurodegenerative disease associated with the death of dopaminergic neurons of the nigrostriatal system. With an increase in life expectancy, the likelihood of developing PD increases. It is known that the death of neurons of the substantia nigra of the brain in this disease occurs due to insufficiency or defection of cellular proteostasis, namely, the process of autophagy, the ubiquitin-proteosome system and the system of chaperone proteins. At present, the etiology of PD is not fully understood, and as a result, existing approaches to the treatment of PD are ineffective. As for most pathologies, an important aspect of ensuring the effectiveness of therapy for PD is diagnostic. Currently, the diagnosis of PD is based on anamnesis and analysis of the clinical picture of the disease. Clinical manifestations of PD occur already in the late stages of the disease, which also explains the ineffectiveness of therapy. Therefore, it is extremely important to identify clinically significant and reliable biomarkers that can predict the progression of PD long before the onset of motor symptoms. Judging by the literature, an active study of the relationship between the various characteristics of peripheral immune system cells and blood plasma with the development of PD is currently underway, but the results obtained by different teams are rather contradictory. This project is aimed at a comprehensive study of the proteostasis system in the cells of the peripheral immune system to identify significant and stable indicators, with a view to their further application in the diagnosis of PD in the early stages
